基于线粒体DNA D-Loop区分析我国沿海厚壳贻贝群体遗传结构

摘要 基于野生厚壳贻贝的线粒体DNA D-Loop区,对我国沿海6个群体(青岛、舟山、玉环、台山、平潭、厦门)的129个厚壳贻贝个体进行遗传多样性及遗传结构分析,获得1113 bp的同源序列,共检测到31个变异位点,得到35个单倍型。A+T含量(64.2%)显著高于G+T含量(35.8%),具有明显的碱基组成偏倚性。结果显示出较高的遗传多样性水平,单倍型多样性(h)为0.853~0.912,核苷酸多样性为(π)为0.00555~0.00736。群体之间的FST值(-0.02179~0.07630)水平较低,差异几乎不显著;通过Nm值(3.02654~32.42974)发现,基因均质化现象明显,基因流导致种群间分化程度甚小。系统进化树显示舟山群体与其他种群有分化现象,但另外5个地区种群的分化程度不高,AMOVA结果显示种群分化都来自于种内,种群间几乎没有遗传分化。单倍型网络图显示6个群体的单倍型没有明显的单支分化。 Genetic diversity and genetic structure of six populations of wild Mytilus coruscus(Qingdao,Zhoushan, Yuhuan, Taishan, Pingtan and Xiamen) in the coast of China were analyzed by amplifying and sequencing the mitochondrial control region(D-Loop). A total of 31 variable sites and 35 haplotypes were detected in 1 113 bp sites for 129 D-Loop sequences. Sequences were analyzed, and the results indicated that the A+T content(64.2%) was higher than that of C+G(35.8%). Population genetic diversity analysis showed that the haplotype diversity(h) and nucleotide diversity(π) of each population were 0.853-0.912 and 0.005 55-0.007 36, respectively. The FSTvalue ranged from-0.021 79-0.076 3, showed a low level of genetic variance among 6 populations. AMOVA analysis showed that all the genetic difference came from within populations.Cluster analysis and haplotype network map showed that the differentiations among populations were not obvious, and the results were consistent with the FSTvalues.
