GAL/GALR2促进涎腺腺样囊性癌细胞神经侵袭的研究

摘要 目的:探究甘丙肽(Galanin,GAL)和甘丙肽2型受体(GALR2)在涎腺腺样囊性癌(SACC)神经侵袭中的作用。方法:将SACC细胞与小鼠背根神经节(DRG)共培养。qRT-PCR、Western Blot检测SACC细胞GALR2的表达;CCK-8、流式细胞术、划痕、Transwell检测GAL对SACC细胞增殖、凋亡及迁移侵袭的影响;神经侵袭实验检测GALR2阻滞(M871)对SACC细胞神经侵袭的影响。结果:共培养SACC细胞GALR2表达显著升高(P<0.05);GAL促进了SACC细胞增殖、迁移侵袭,抑制凋亡(P<0.05);M871可抑制SACC细胞神经侵袭。结论:神经来源GAL可激活SACC细胞的GALR2,促进其神经侵袭。 Objective:To explore the role of galanin(GAL)and galanin receptor 2(GALR2)in perineural invasion(PNI)of salivary adenoid cystic carcinoma(SACC)cells.Methods:SACC cells were co-cultured with dorsal root ganglion(DRG).qRT-PCR and Western Blot were used to detect GALR2 expression in SACC cells.The effects of GAL on the proliferation apoptosis,migration and invasion of SACC cells were detected by CCK-8 assay,flow cytometry,wound healing scratch,and Transwell assays.The effect of GALR2 blocking M871 on PNI of SACC cells was detected by nerve invasion assay.Results:The expression of GALR2 in co-cultured SACC cells was increased(P<0.05).GAL promoted the proliferation,migration and invasion but inhibited the apoptosis of SACC cells(P<0.05).M871 inhibited the PNI of SACC cells.Conclusion:Neurogenic GAL may activate the expression of GALR2 in SACC cells and promote the PNI of the cells.
