复方中草药制剂抑菌作用及其对湖羊部分细胞因子mRNA表达的影响

摘要 【目的】检测复方中草药制剂对引起湖羊乳房炎的金黄色葡萄球菌、大肠杆菌和粪肠球菌的抑杀作用,并探究该制剂对湖羊外周血中部分细胞因子mRNA相对表达量的影响.【方法】采用牛津杯法和试管二倍稀释法进行复方中草药制剂的体外抑菌试验,荧光定量检测用药前和用药后第1、3、7、14天及21天湖羊外周血中IL-2、IL-6、IL-8、IL-10、IL-12、IFN-γ和TNF-α等7种细胞因子mRNA相对表达量.【结果】体外抑菌试验结果显示,该复方中草药制剂对大肠杆菌、粪肠球菌和金黄色葡萄球菌最小抑菌浓度(MIC)分别达到125、125、62.5 mg/mL;最小杀菌浓度(MBC)分别为250、125、125 mg/mL.荧光定量试验结果显示,使用该制剂处理湖羊后能显著提高湖羊外周血中IL-2、IL-8、IL-10、IL-12和IFN-γ的mRNA表达水平(P<0.05),显著降低IL-6和TNF-αmRNA表达水平(P<0.05).【结论】该复方中草药制剂对3种乳房炎病原菌具有很好的抑杀作用,并能上调湖羊外周血中IL-2、IL-8、IL-10、IL-12和IFN-γ的mRNA相对表达量,下调IL-6和TNF-α的mRNA相对表达量. 【Objective】To detect the antibacterial effect of the compound Chinese herbal medicine preparations on the pathogens of Hu sheep mastitis including Staphylococcus aureus,Escherichia coli and Enterococcus faecalis,and explore the effect of the preparation on the relative mRNA expression of cytokines in peripheral blood of Hu sheep.【Method】In vitro bacteriostasis experiment of compound Chinese herbal medicine preparation was carried out by Oxford cup method and double dilution method,the mRNA expression levels of IL-2,IL-6,IL-8,IL-10,IL-12,IFN-γand TNF-αin peripheral blood of Hu sheep,were measured by quantitative real-time PCR test,before and after medication 1 d,3 d,7 d,14 d and 21 d.【Result】The results of in vitro antibacterial tests showed that the minimum inhibitory concentration(MIC)of the compound Chinese herbal medicine preparation against E.coli,E.faecalis and S.aureus respectively reached 125 mg/mL,125 mg/mL and 62.5 mg/mL.The minimum bactericidal concentration(MBC)respectively reached 250 mg/mL,125 mg/mL and 125 mg/mL.Quantitative real-time PCR test results showed that the preparation could significantly increase the mRNA expression levels of IL-2,IL-8,IL-10,IL-12,and IFN-γ,significantly reduced mRNA expression levels of IL-6 and TNF-α,in peripheral blood of Hu sheep.【Conclusion】The compound Chinese herbal medicine preparation has a significantly antibacterial effect on three kinds of mastitis pathogens,and it can up-regulate the mRNA expression of IL-2,IL-8,IL-10,IL-12 and IFN-γ,down-regulate the mRNA expression of IL-6 and TNF-α,in peripheral blood of Hu sheep.
