藏药佐太和甲基汞对神经细胞Neuro-2a毒性的差异比较

摘要 【目的】探索藏药佐太和甲基汞对神经细胞Neuro-2a凋亡情况的影响差异.【方法】用磺基罗丹明B法测定细胞存活率,HE染色观察细胞形态变化,流式细胞术检测细胞凋亡率,实时荧光定量PCR检测凋亡相关基因表达量的变化.【结果】与空白组相比,随着孵育时间的增加(6、12、24 h),佐太组(1.83 mg/L,含汞1 mg/L)的细胞存活率和细胞形态无显著变化,孵育12 h时细胞凋亡率显著增加,孵育24 h时炎症因子基因IL-6的表达显著增加,对Caspase家族基因和线粒体相关基因的表达影响无显著影响;而甲基汞组(1.25 mg/L,含汞1 mg/L)的细胞存活率由(97.1±0.082)%降至(26.3±0.069)%,细胞形态被破坏程度逐渐加剧,细胞凋亡率随孵育时间的增加而显著增加,且均高于佐太组,炎症因子基因IL-6、IL-1β、TNF-α的表达显著增加,线粒体凋亡相关基因Bcl-2和Bax的比值表达降低,Caspase-3和Caspase-9基因的表达增加.【结论】汞含量相同时藏药佐太对神经细胞Neuro-2a的毒性远小于甲基汞. 【Objective】To explore the differences between Tibetan medicine Zuotai and methyl mercury on the apoptosis of Neuro-2a cells.【Method】The cell activity was determined by SRB method.The haematoxylin and eosin(HE)was used to observe the morphology of Neuro-2a cells.The apoptosis rate was detected by Amnis Flow Sight System.Real-time PCR was used to examine the expressions of the apoptosis-related genes.【Result】Compared with the control group,with the increase of incubation time(6,12,24 h),no significant changes in cell viability and cell morphology were found in Zuotai group(1.83 mg/L,the mercury content was 1 mg/L),the apoptosis rate increased significantly after incubation for 12 h,the expression of inflammatory factor IL-6 was significantly increased in Zuotai group after incubation for 24 h,and there were no significant differences in the expression of Caspase family genes and mitochondria-related genes.The cell viability of MeHgCl group(1.25 mg/L,the mercury content was 1 mg/L)was decreased from(97.1±0.082)%to(26.3±0.069)%,the morphology was gradually seriously damaged,and the apoptosis rate of MeHgCl group was increased with the increase of incubation time,which were higher than that of Zuotai group.The expression of inflammatory cytokine genes IL-6,IL-1βand TNF-αwas significantly increased in MeHgCl group,the ratio of mitochondrial apoptosis-related genes between Bcl-2 and Bax was decreased,and increased the expression of Caspase family genes Caspase-3 and Caspase-9.【Conclusion】The toxicity of Tibetan Zuotai to Neuro-2a is much less than that of methyl mercury at same mercury content.
