高原线虫草子座、菌核及生境土壤真菌多样性分析

摘要 采用Illumina Miseq高通量测序技术对高原线虫草(Ophiocordyceps highlandensis)子座、菌核和生境土壤样本真菌ITS序列进行测定,分析其真菌多样性和群落结构。所测样本共获得1650个OTU,子座、菌核和生境土壤的OTU数量分别为524、272和854个,注释的真菌隶属于19纲70目132科161属。子座和菌核组间、子座和生境土壤组间真菌多样性无显著性差异;菌核和生境土壤组间真菌多样性具有显著性差异,子座和菌核组间真菌丰富度无显著性差异,子座和生境土壤组间、菌核和生境土壤组间真菌丰富度具有显著性差异。子囊菌门(Ascomycota)为子座、菌核和生境土壤真菌群落的最大优势门,子座中的优势属为Circinotrichum、轮枝孢菌属(Verticillium)和多头霉属(Polycephalomyces),菌核中的优势属为泛普可尼亚菌属(metapochonia)、地丝霉属(Geomyces)和轮枝孢菌属(Verticillium),生境土壤中的优势属为Cadophora。子座、菌核和生境土壤特有真菌物种分别有33、3、343个,共有真菌物种有237个。该研究为开发利用高原线虫草微生物群落中真菌资源提供参考。 Using the lllumina Miseq high-throughput sequencing technology,fungal ITS sequences in stroma,sclerotium and habitat soil of Ophiocordyceps highlandensis were sequenced and then analyzed for fugal diversity and community structure.The results showed that a total of 1650 OTUs were identified in the tested samples,including 524 OTUs in stroma,272 OTUs in sclerotium and 854 OTUs in the habitat soil,respectively.The annotated sequences belonged to 161 genera,132 families,70 orders,and 19 classes.There was no significant difference in fungal diversity between stroma and sclerotium or between stroma and the habitat soil.There was a significant difference in fungal diversity between sclerotium and the habitat soil.No significant difference in fungal abundance was found between stroma and sclerotium,but fungal abundance in the habitat soil was significantly different from that in either stroma or sclerotium.Ascomycota was the dominant phylum of fungi in stroma,sclerotium and the habitat soil.Dominant genera in stroma were Circinotrichum,Verticillium and Polycephalomyces;dominant genera in sclerotium were metapochonia,Geomyces and Verticillium;and the dominant genus in the habitat soil was Cadophora.Venn diagram showed that there were 33,3,and 343 fungal species endemic to stroma,sclerotium and the habitat soil,respectively;and 237 mutual fungal species.These findings provide a reference for the development and utilization of fungal resources in Ophiocordyceps highlandensis.
