旱莲苷对阿尔茨海默病大鼠学习记忆的影响

摘要 目的研究旱莲苷对阿尔茨海默病(AD)大鼠学习记忆及海马组织乙酰胆碱酯酶(AchE)、糖原合酶激酶3β(GSK-3β)、乙酰胆碱转移酶(ChAT)的影响。方法将60只大鼠随机分为对照组(n=20)、模型组(n=20)及实验组(n=20)。实验组及模型组大鼠均用腹腔注射D-半乳糖联合双侧海马注射β-淀粉样蛋白(Aβ)25-35构建AD大鼠模型,对照组大鼠注射等量生理盐水。造模后,实验组大鼠灌胃24 g·kg-1·d-1旱莲苷,模型组与对照组灌胃等量生理盐水,连续干预8周。用Morris水迷宫检测大鼠空间学习记忆能力;用免疫组化法及蛋白质印迹(Wb)法检测海马组织AchE、GSK-3β、ChAT蛋白表达。结果对照组、模型组及实验组大鼠第4天逃逸潜伏期分别为(9.82±3.06),(50.69±9.51)和(26.87±8.12)s;首次到达原平台位置的时间分别为(15.42±1.31),(41.12±20.34)和(22.76±5.87)s;穿越原平台次数分别为(5.39±2.01),(1.53±1.21)和(3.36±0.58)次;海马组织AchE蛋白相对表达量分别为0.23±0.11,0.96±0.04和0.47±0.03;GSK-3β蛋白相对表达量分别为0.28±0.13,0.98±0.02和0.41±0.04;ChAT蛋白相对表达量分别为0.95±0.05,0.46±0.12和0.84±0.09。以上指标:模型组与对照组比较,差异均有统计学意义(均P<0.05);实验组与模型组比较,差异均有统计学意义(均P<0.05)。模型组大鼠海马组织AchE、GSK-3β蛋白阳性细胞百分比均高于对照组,与模型组比,实验组以上指标均降低;而ChAT蛋白阳性细胞百分比降低,且实验组高于模型组(均P<0.05)。结论旱莲苷可改善AD大鼠海马组织病变,提高其学习记忆能力,作用机制可能与下调海马组织AchE、GSK-3β蛋白表达,上调ChAT蛋白表达有关。 Objective To investigate the effects of lycopene on learning and memory and acetylcholinesterase(AchE),glycogen synthase kinase 3β(GSK-3β)and acetylcholine transferase(ChAT)in hippocampus of Alzheimer’s disease(AD)rats.Methods Sixty rats were randomly divided into control group(n=20),model group(n=20)and test group(n=20).Rats in model group and test group were treated with intraperitoneal injection of D-galactose and bilateral hippocampal injection ofβ-amyloid protein(Aβ)25-35 to construct the AD rat model,rats in control group were injected with the same amount of normal saline.After successful modeling,rats in test group were given 24 g·kg-1·d-1 lycopene,rats in model group and control group were given the same amount of normal saline for 8 weeks.The spatial learning and memory ability of rats were detected by Morris water maze tester;the expression of AchE,GSK-3βand ChAT protein in hippocampus were detected by immunohistochemistry and Western blotting(Wb)method.Results The escape latency of control group,model group and test group on the 4thd were(9.82±3.06),(50.69±9.51)and(26.87±8.12)s;the time when reached the original platform position for the first time were(15.42±1.31),(41.12±20.34)and(22.76±5.87)s;the number of crossing the original platform were(5.39±2.01),(1.53±1.21)and(3.36±0.58)times;the relative expression levels of AchE protein in hippocampus were 0.23±0.11,0.96±0.04 and 0.47±0.03;the relative expression levels of GSK-3βprotein were 0.28±0.13,0.98±0.02 and 0.41±0.04;the relative expression levels of ChAT protein were 0.95±0.05,0.46±0.12 and 0.84±0.09.Comparing between model group and control group,the differences of the factors were significant(all P<0.05);comparing between test group and model group,the differences of the factors were significant(all P<0.05).Compared with control group,the percentage positive cells of AchE and GSK-3βprotein in hippocampus of model group were significantly increased,and test group was lower than model group;the percentage positive
